Dear all,

Following what we discussed in the last Proteored meetings, we are going to launch a new multi-centric experiment, PME6. The main goal this time is going to be to analyze by LC-MS/MS a medium complexity sample, that can be then useful as a standard to check performance of instruments, protocols,...
The sample has been prepared by the PCB lab, by Lia and her colleagues, and you will receive it during this week. The sample will be delivered frozen, in a low-binding eppendorf in 37.5 ul 50mM Tris-NaCl pH 7.4. There should be 50 to 100 ug of protein in your eppendorf.
The sample consists in a mixture of plasma proteins and some standard spiked-in proteins. What we propose is to analyze this sample on a single LC-MS/MS run, which will allow us to compare the performance of the procedure by checking for instance:

  • number of proteins and peptides identified
  • specificity and sensitivity of the protein identification
  • attained MS dynamic range

We will send later this month details about the sample and some directions to be followed for the analysis and reporting of the results.
